Cocos nucifera (old coconut): an alternative medium for nutrient agar.


One of the important problems in microbiology laboratories is an expensive culture media that can grow many types of microorganisms depending on the research needs. One of which that is commonly used is an animal-based media known us Nutrient agar. The study is about the potential of this mature coconut water we generally dispose upon cracking the coconut shells. The researchers come up to an idea to make a culture media out of plant that can supply most of the nutrient requirement of the bacteria.

Cocos nucifera (old coconut) produced locally was tested for its ability to support the growth of Staphyloccus aureus ATCC-25923, Bacillus subtilis, and Escherichia coli ATCC-25922 form University of the Philippines, Manila. All glass wares were autoclaved at 121°C, 15 psi to avoid contamination. To solidify the medium use 8 grams of gelatin powder per 100 ml solution. From 5 Mcfarland, bacteria were diluted into 1/1000 dilution. 1/1000 bacteria were inoculated using streak plate method on agar plates. Fifteen percent (15%) concentration of mature coconut water produces a significant growth of colonies within 24 hours of incubation counted in cfu/uL. Mature coconut water based culture medium could therefore be an alternative medium in terms of supporting the growth of bacteria for nutrient agar.
