Acid-fast microscopy in respiratory tuberculosis.

Sputum of one hundred (100) subjects diagnosed radiologically as respiratory tuberculosis were subjected to direct smear microscopy to detect acid-fact organisms using Ziehl-Neelsen and Kinyoun stain method to determine the difference in the percentage of yield under local laboratory practice. Direct smear microscopy of early morning sputum specimen subjected to both staining methods revealed 19 patients positive for acid-fast bacilli with Ziehl-Neelsen stain and 31 patients positive with Kinyoun stain method. The finding of a relatively large number of cases positive with Kinyoun stain methods in this study showed that the use of the said procedure is more likely to yield a positive result compared to Ziehl-Neelsen stain method (p-value=0.0001).
