Objectives

General Objective: To develop a method for the rapid determination of resistance to INH exhibited by M. tuberculosis isolates.Specific Objectives:1. To determine which of the existing probes and primers should be used for the amplication and recognition of the Kat G gene of both sensitive and resistant isolates;2. To determine a gold standard for a Kat G. gene of INH sensitive isolates;3. To determine the deviations of Kat G. genes from resistant isolates from this standard;4. To correlate to degree of resistance to INH and the type of mutation observed; and5. To determine if the gene amplification and sequencing techniques developed may be used to determine resistance of sensitivity of isolates in sputum samples.
