Determination of the anti-angiogenic activity of Cinnamomum cebuense through chorioallantoic membrane (CAM) assay.

Background: Vascular endothelial growth factor (VEGF) has become an opportune therapeutic target for anti-cancer drugs as it has been implicated in tumor angiogenesis. Antiangiogenic drugs are limited by the side effects that accompany their chronic use; these include hypertension, bleeding, and GI perforation. A potential alternative therapeutic approach is the identification of naturally occurring diet derived angiogenesis inhibitors. Polyphenols from Cinnamon have recently been recognized for its ability to inhibit VEGF induced signaling in endothelial cells. The proposed study aimed to evaluate the antiangiogenic activity of Cinnamomum cebuense extract, a Cinnamon species endemic to the forest fragments of Cebu, Philippines. Elucidation of its novel bioactive property may aid in the preservation of the critically endangered species. Methods: Air dried Cinnamon bark was separated into its liquid component through boiling, decantation, and centrifugation. The resultant supernatant was then been transferred to sterile test tubes for dilution with distilled water and refrigerated at 10C. Meanwhile, fertile day 0 mallard duck eggs were incubated at 37C. On day 4, albumin was removed and on Day 8, 40 µL of the concentrations of each sample (10%, 25%, 50% and 75% cinnamon extract, 100% Nexavar) was added directly onto the CAM. The eggs were then returned to the incubator for another 72 hours. A stereomicroscope was utilized to visually assess the vascular damage. Photographs taken of viable eggs were analyzed with Image J, a software used to measure blood vessel density and quantify blood vessel tributaries, thereby quantifying antiangiogenic activity. Data gathered was analyzed using one sample T-Test and Independent T-Test. Results: Statistical analysis revealed that Cinnamomum cebuense at concentrations of 50% and 70% were found to have significant effects in decreasing the number of primary blood vessels. In addition, 50% Cinnamomum also had a lowering effect for both secondary and tertiary blood vessels. Conclusion: Cinnamomum cebuense possesses anti-angiogenic activity.
