Duhat bark collected from Meycauayan, Bulacan was used in this research for its tannin constituents. Three types of samples were prepared for extraction - fresh, sundried, and airdried. Different methods of extraction namely decoction, infusion, maceration and percolation using alcohol as menstrum besides water separately in the last two methods. Aqueous extracts were concentrated by direct heating and alcoholic extracts by water bath. Among the concentrated extracts obtained, percolation of sundried sample using alcohol as menstrum showed the highest percentage yield of tannins. The product had a reddish brown color. Pharmaceutical books gave a similar color of product. The percolation of sundried sample using alcohol as menstrum was used as the tannin extract for the formulation of mouthwash. This tannin extract and citric acid were weighed and dissolved in distilled water. The solution was set aside while peppermint spirit and methyl salicylate were dissolved in alcohol. This alcoholic solution was added to the aqueous solution and filtered to obtain clear product. A sufficient quantity of distilled water was added through the filter paper making the product measure 60 ml and transferred in 60 ml amber colored bottle and stoppered securely to protect from light. The tannerine mouthwash was tested for acceptability by thirty members of the test panel. Based on the evaluation of the test panel, the mouthwash proved to be acceptable in terms of its odor, taste, color and viscocity. Based on microbiological testing using the paper disc method, the tannerine mouthwash was found to have antiseptic property inhibiting the growth of Streptococcus mutans which is a gram (+) microorganism. It is found to be also inhibitive against Bacteriodas melaninogenicus (gram(-) microorganism) to a lesser extent. Thus, tannerine mouthwash is more potent against Streptococcus mutans (gram(-) microorganism) which is a resident bacteria in the mouth.
The study aimed to look into the best possible way to extract tannin, todetermine which gives the higher tannin percentage yield content, the fresh, the air dried or the sundried bark and to know how isit possible to formulate mouthwash preparation.
